Roles of Heparin-Binding Protein in Bacterial Infections

چکیده
Infectious diseases remain a major health problem, where sepsis and other severe infectious diseases are common causes of morbidity and mortality. The importance of early and appropriate treatment of sepsis and severe bacterial infections has been underlined by the successes of measures like the Surviving Sepsis Campaign, among others. Thus, there is a need for clinical and laboratory tools to identify a patient with severe infection early and to distinguish between bacterial and non-bacterial conditions. Heparin-binding protein (HBP) is also called azurocidin, or cationic antimicrobial protein of 37 kDa (CAP37). It is a multifunctional granule-associated protein that is rapidly mobilized from migrating polymorphonuclear leukocytes. HBP acts as a chemoattractant, an activator of monocytes and macrophages, and induces vascular leakage and edema formation. The release of HBP is triggered by ligation of neutrophilic 2 -integrins, a process that may be initiated by bacterial structures. The overall outcome is powerful vascular leakage. It has been shown that patients with severe sepsis express high levels of HBP in plasma before they develop hypotension. HBP is also involved in the pathophysiology of soft tissue inReceived: February 11, 2010 Accepted after revision: March 20, 2010 Published online: May 12, 2010 Journal of Innate Immunity Dr. Adam Linder Department of Clinical Sciences, Division of Infection Medicine Lund University Hospital SE–221 85 Lund (Sweden) Tel. +46 46 171 309, Fax +46 46 323 895, E-Mail adam.linder @ skane.se © 2010 S. متن کاملHSP Roles as Biomarkers and Antigens in Bacterial and Viral Infections
Diagnosis of infectious diseases remains an important issue in medical science. Identification of biomarkers can be used to predict early infections. Recently, heat shock proteins (HSPs) have been known as the conserved compounds expressed under stress conditions in both prokaryotic and eukaryotic systems. These proteins act as molecular chaperones. Several studies showed the increased levels o...
متن کاملImmunogenicity of heparin-binding hemagglutinin expressed by Pichia pastoris GS115 strain
Objective(s): Heparin-binding hemagglutinin (HBHA), a mycobacterial cell surface protein, mediates adhesion to nonphagocytic cells and the dissemination of Mycobacterium tuberculosis (M. tuberculosis) from the site of primary infection. Superior expression systems are required to obtain abundant M. tuberculosis proteins for the purpose of diagnosing M. tuberculosis infection or for the immuniza...
متن کاملM Protein, a Classical Bacterial Virulence Determinant, Forms Complexes with Fibrinogen that Induce Vascular Leakage
Increased vascular permeability is a key feature of inflammatory conditions. In severe infections, leakage of plasma from the vasculature induces a life-threatening hypotension. Streptococcus pyogenes, a major human bacterial pathogen, causes a toxic shock syndrome (STSS) characterized by excessive plasma leakage and multi-organ failure. Here we find that M protein, released from the streptococ...
متن کاملNovel interactions of glycosaminoglycans and bacterial glycolipids mediate binding of enterococci to human cells.
Enterococcus faecalis is among the most important nosocomial pathogens. The intestinal mucosa is considered to be the main site used by these bacteria for entrance and dissemination. A better understanding of the mechanisms involved in colonization and invasion of enterococci may help to devise methods to prevent infections in hospitalized patients. Glycosaminoglycans, which are present on the ...
